چکیده

Molecular analysis of a mutant wary allele, wx-mum5 (Don that of Mul (67%), but significantly higher than the G/C content Robertson, unpublished), revealed the presence of a previously of the entire maize genome, estimated to be 49.5% G/C (2). unidentified maize Mutator element (Mu8) within the tenth exon of the waxy gene. The element is 1410 base pairs (bp) long and is flanked by a nine bp direct repeat of insertion site DNA. The element has inverted terminal repeats of 185 bp which are 96.8% REFERENCES homologous (179/185). These terminal repeats are 89.2% and Figure 1. The complete nucleotide sequence of Mu8. The solid line beneath the sequence shows the extent of the inverted terminal repeats which are flanked by the 9 base pair direct repeat of insertion site DNA.

Identification of an Active New Mutator Transposable Element in Maize

Robertson's Mutator (Mu) system has been used in large scale mutagenesis in maize, exploiting its high mutation frequency, controllability, preferential insertion in genes, and independence of donor location. Eight Mutator elements have been fully characterized (Mu1, Mu2 /Mu1.7, Mu3, Mu4, Mu5, Mu6/7, Mu8, MuDR), and three are defined by TIR (Mu10, Mu11 and Mu12). The genome sequencing revealed ...

Sequence analysis of the cloned glossy8 gene of maize suggests that it may code for a beta-ketoacyl reductase required for the biosynthesis of cuticular waxes.

The gl8 locus of maize (Zea mays L.) was previously defined by a mutation that reduces the amount and alters the composition of seedling cuticular waxes. Sixty independently derived gl8 mutant alleles were isolated from stocks that carried the Mutator transposon system. A DNA fragment that contains a Mu8 transposon and that co-segregates with one of these alleles, gl8-Mu3142, was identified and...

Sequence similarity of putative transposases links the maize Mutator autonomous element and a group of bacterial insertion sequences

The Mutator transposable element system of maize is the most active transposable element system characterized in higher plants. While Mutator has been used to generate and tag thousands of new maize mutants, the mechanism and regulation of its transposition are poorly understood. The Mutator autonomous element, MuDR, encodes two proteins: MURA and MURB. We have detected an amino acid sequence m...

Sequence, genomic distribution and DNA modification of a Mu1 element from non-mutator maize stocks.

The increased mutation rate of Mutator stocks of maize has been shown to be the result of transposition of Mu elements. One element, Mu1, is present in 10-60 copies in Mutator stocks and approximately 0-3 copies in non-Mutator stocks. The sequence, structure and genomic distribution of an intact Mu1 element cloned from the non-Mutator inbred line B37 has been determined. The sequence of this el...

Characterization of a highly conserved sequence related to mutator transposable elements in maize.

Mutator stocks of maize exhibit a high mutation rate correlated with the activity of a family of transposable elements. Mu1 and, to a lesser extent, the closely related Mu1.7 elements are responsible for most mutator-induced mutations that have been characterized. These elements are found in 10-60 copies in mutator stocks, and zero to a few intact elements exist in nonmutator maize stocks. Addi...
